1. The shotgun sequencing run of Lentinula edodes generates 115 Mbp data with an average of 250bp.

2. Emulsion PCR breaking and DNA beads enrichment were done for Pleurotus cDNA library (16 x 3 cpb and 16 x 3.5 cpb). The % good DNA beads recovery is a bit lower than expected and 6 additional emPCR is necessary.

1. LR 70 Sequencing run for Lentinula edodes was started today and approximately 850,000 beads were loaded per region.
2. For Pleurotus cDNA library, 32 emulsion PCR were done today( 3 cpb x 16 and 3.5 cpb x 16) and will be broken tomorrow for recovery.

Results
It was good that 10% enrichment was reached and there would be sufficient beads to be loaded on the PTP.

Others
1. The tendering procedures for the HK$1M titanium reagents were completed and Roche had received the order on Dec 22.
2. Platinum High-Fidelity Taq was ordered and will be delivered on Dec 29.
3. The installation and calibration of the ABI 7500 RealTime PCR machine was finished. A training couse will be arranged after the holiday. Let me know if you want to join.
